Proteomic Analysis of Mesenchymal Stromal Cells Secretome in Comparison to Leukocyte- and Platelet-Rich Fibrin

Secretomes of mesenchymal stromal cells (MSCs) are emerging as a novel growth factor (GF)-based strategy for periodontal and bone regeneration. The objective this study was to compare the secretome human marrow MSC (BMSC) that leukocyte- platelet-rich fibrin (L-PRF), an established GF-based therapy, in context wound healing Conditioned media from BMSCs (BMSC-CM) L-PRF (LPRF-CM) were subjected quantitative proteomic analysis using liquid chromatography with tandem mass spectrometry. Global profiles, gene ontology (GO) categories, differentially expressed proteins (DEPs), set enrichment (GSEA) identified bioinformatic methods. Concentrations selected determined multiplex immunoassay. Among BMSC-CM (2157 proteins) LPRF-CM (1420 proteins), 1283 common. GO revealed similarities between groups terms biological processes (cellular organization, protein metabolism) molecular functions (cellular/protein-binding). Notably, more DEPs (n = 550) compared 118); these included several key GF, cytokines, extracellular matrix (ECM) involved healing. GSEA ECM (especially ECM)-related immune-related LPRF-CM. Similar trends intergroup differences detection observed analysis. Thus, BMSC is enriched proteins/processes relevant vivo efficacy therapy should be evaluated future studies.
